In Vivo Microdialysis Study for the Effect of Repeated Nicotine Treatment on the Brain Dopamine and Serotonin Metabolism


Abstract
Nicotine is one of the most widely used psychotropic drugs and has important effects on brain dopaminergic neurotransmission. The effects of repeated treatment with nicotine (1.5mg/kg. s.c.) on 3. 4-dihydroxyphenylaeetic acid (DOPAC) turnover of
the rat
striatum and nucleus accumbens was studied using in vivo microdilysis measurement of extracellualar concentrations of DOPAC. 5-hydroxyindoleacetic acid (5-HlAA) and homovanilic acid (HVA) in urethane anesthetised ratas and also the acute nicotine
effect
on dopamine metabolism was evaluated in the rats given with repeated nicotine treatment for 2 weeks. Basal extracellular level of DOPAC in nucleus accumbens was decreased with repeated nicotine petreatment. but increase of extracellular level of
DOPAC
in nucleus accumbens by nicotine was not significantly altered by repeated treatment of the nicotine. The increase of extracellular level of 5-HlAA in nucleus accumbens in response to acute nicotine injection was potentiated by repeated nicotine
treatment. The extracellular DOPAC turnover measured after administration of pargyline were decreased in nucleus accumbens by repeated nicotine treatment. These results suggested that basal extracellular DOPAC and its turnover in nucleus
accumberns
were
decreased by repeated nicotine pretreatment but the increase of extracellular DOPAC induced by acute nicotine injection was not altered by repeated nicotine pretreatment.
